A micromethod for the determination of ferroxidase (ceruloplasmin) in human serums.

چکیده
A sensitivemethod for the detection of ceruloplasmin (ferroxidase) in human serums, using Fe(Il) as the substrate, is described. Initial Fe(ll) concentrations of 30 and 120 p.M may be used, depending on the sensitivity of the spectrophotometer, for the determination of the rate of Fe(Ill) -transferrin formation following the oxidation of Fe(lI) by ceruloplasmin (ferroxidase). Advantages of this method over other methods employing p-phenylenediamine as a substrate and the standardization of enzymeactivityin termsof international enzyme units are discussed.

Automated bichromatic analysis of serum ceruloplasmin.
An automated micromethod is described for measurement of serum ceruloplasmin by assay of its p-phenylenediamine oxidase activity using the Abbott bichoromatic analyzer. Ten mul of serum are mixed with 250 mul of p-phenylenediamine (PPD) substrate (9.2 mmole per liter) in acetate buffer (0.1 mole per liter, pH 5.45). Spectrophotometric measurements of the rate of formation of the purple oxidatio...

In the blood plasma of humans and rats, ceruloplasmin is the major copper-binding protein and ferroxidase, accounting for 70% of the copper present in the plasma, with the rest binding primarily to albumin and a macroglobulin. Systematic studies with fresh plasma were carried out to compare what occurs in the mouse. C57BL6 mice had half as much copper and pPD (p-phenylene diamine) oxidase activ...

Physiol. 227(5) : 1094-1097. 1974.-The ferroxidase activity of rat ceruloplasmin was studied under various assay conditions. The ferroxidase activity was optimal at pH 6.0-6.2 in acetate buffer in the absence of ascorbate, and under these conditions the activity was about 1/3rd that of human cer...
متن کاملFunctional relevance of ceruloplasmin mutations in Parkinson's disease.
Increased iron levels of the substantia nigra and the discovery of ceruloplasmin mutations in patients with Parkinson's disease (PD) imply impaired iron metabolism in this neurodegenerative disorder. Ceruloplasmin has ferroxidase activity oxidizing iron(II) to iron(III). In the present study, we analyzed the amount of ceruloplasmin, iron, ferritin, and transferrin and the ceruloplasmin ferroxid...
متن کاملAutomated measurement of serum ferroxidase activity.
A method is described for automated measurement of serum ceruloplasmin ferroxidase activity. In this method, Fe2+ ions are used as the substrate. In addition, a new calibration system without ceruloplasmin is also presented. Optimum assay reaction conditions were determined. Maximal catalytic activity was obtained at 0.45 mol/L acetate buffer, pH 5.8. The reagents and calibrator are stable for ...
